We appreciate the thoughtful engagement of Dalgleish et al regarding the interaction between Human Genome Variation Society (HGVS) Nomenclature and the file system constraints we outlined in our article.1 The authors correctly highlight that specific characters used in canonical HGVS descriptions (eg, :, /, >, ∗) are incompatible with common operating system file and directory naming conventions. They further caution against the risk of propagating alternative character encodings into scientific communication and clinical reporting, and we fully agree with their concerns about a lack of interoperability in this approach.
